Indications
- Amoebiasis

Pharmacology
- Ornidazole is a 5-nitroimidazole derivative active against protozoa and anaerobic bacteria
- Interacts with DNA to cause destruction of helical DNA structure
Dosage & Administration
- Adult dosage for amoebiasis: 500 mg twice a day for 5 days
- Children dosage for amoebiasis: 10-25 mg per kg body weight in two divided doses
- Adult dosage for amoebic dysentery: 1.5 gm once a day for 3 days
- Children dosage for amoebic dysentery: 40 mg per kg body weight, once a day for 3 days
- Adult dosage for giardiasis: 1.5 gm once daily for 1-2 days
- Children dosage for giardiasis: 40 mg per kg body weight for 2 days
- Adult dosage for trichomoniasis: 1.5 gm once or 500 mg twice a day for 5 days
- Treatment of bacterial vaginosis: 3 tablets of 500 mg each as a single dose or one tablet of 500 mg once daily for 5-7 days
Interaction
Mild potential to cause disulfiram-like reactions, may increase the risk of hemorrhage due to diminished hepatic metabolism, reported to decrease the clearance of 5-fluorouracil
Contraindications
- Previous hypersensitivity to Ornidazole and nitroimidazoles
- Contraindicated in central nervous system disorders, epilepsy, or peripheral neuropathy
Side Effects
- Gastrointestinal tract effects: nausea, vomiting, epigastric pain
- Central nervous system effects: dizziness, headache, lassitude
- Leukopenia, headache, dizziness, lassitude, fatigue, weakness
Pregnancy & Lactation
Adequate clinical trials have not been conducted. Ornidazole should be prescribed only if the potential benefit justifies the potential risk to fetus/neonate
Precautions & Warnings
- Prescribed with caution in patients with ataxia, vertigo, and mental confusion
- Rarely reported blood dyscrasia: mild leukopenia
- Discontinue therapy if leukopenia occurs, based on the gravity of infection
Use in Special Populations
- Renal Impairment: Supplemental dose for haemodialysis patients
- Hepatic Impairment: Double the interval between doses for severe cases
