Indications

Composition
- Each film coated tablet contains Chloroquine Phosphate BP 250 mg eqv. to approx. 150 mg of Chloroquine base
- Each 5 mL syrup contains Chloroquine Phosphate BP 80 mg eqv. to approx. 50 mg of Chloroquine base
Pharmacology
- 4-aminoquinoline antimalarial and amoebicidal agent
- Used for the suppression and clinical cure of malaria caused by susceptible strains of Plasmodium species
- Rapidly acting blood schizontocide with gametocytocidal activity
- Mechanism of action may involve influencing haemoglobin digestion and interfering with parasite nucleoprotein synthesis
- Rapid and complete absorption from gastrointestinal tract
- Approximately 55% bound to plasma proteins
Dosage & Administration
- Dosage based on chloroquine base
- Different dosing regimens for acute attack of malaria in adults and children
- Specific prophylaxis doses for adults and children
- Dosage for hepatic amoebiasis, discoid and systemic lupus erythematosus, rheumatoid arthritis, photoallergic reactions, and porphyria cutanea tarda
- Special dosage considerations for children and adults in various conditions
Interaction
- Exhibits interaction with neostigmine, pyridostigmine, antacids, kaolin, cimetidine, ranitidine, quinine, mefloquine, amodiaquine, artemisinin, metronidazole, and ampicillin
Contraindications
- Known or suspected chloroquine resistance in P. falciparum
- Hypersensitivity
- Retinal damage
- Concurrent therapy with hepatotoxic drugs
Side Effects
- Gastro-intestinal: Nausea, vomiting, and diarrhoea
- Psychological: Psychotic episodes, anxiety, and personality changes
- Visual disturbances: Blurred vision, difficulties in focusing, keratopathy, and retinopathy
- Haematological: Aplastic anaemia, agranulocytosis, thrombocytopenia, and neutropenia
- Others: Loss of hair, bleaching of hair pigment, pigmentation of mucous membranes and skin, photosensitivity, tinnitus, reduced hearing, nerve deafness, uromyopathy, and myopathy
Pregnancy & Lactation
Avoid use during pregnancy unless benefit outweighs possible hazard; consider discontinuing nursing if using the drug
Precautions & Warnings
- Examine eyes before starting long-term treatment
- Monitor visual disturbances
- Administer with care to patients with impaired liver or renal function, or with porphyria, psoriasis, or a history of epilepsy
- Observe patients with G6PD deficiency for haemolytic anaemia
