Indications
- Treatment of malaria

Composition
- Tablet: Each film coated tablet contains Chloroquine Phosphate BP 250 mg equiv. to approx. 150 mg of Chloroquine base
- Syrup: Each 5 mL contains Chloroquine Phosphate BP 80 mg equiv. to approx. 50 mg of Chloroquine base
Pharmacology
- Antimalarial and amoebicidal agent
- Suppression and clinical cure of malaria due to susceptible strains of Plasmodium sp.
- Mechanism of action influences haemoglobin digestion and interferes with parasite nucleoprotein synthesis
Dosage & Administration
- Doses expressed in terms of chloroquine base
- Different dosages for acute attack of malaria, clinical prophylaxis, hepatic amoebiasis, discoid and systemic lupus erythematosus, rheumatoid arthritis, management of photoallergic reactions, porphyria cutanea tarda
- Dosage expressed for adults and children
- Specific dosages for different conditions
Interaction
- Exhibits interaction with drugs like neostigmine, pyridostigmine, antacids, kaolin, cimetidine, ranitidine, quinine, mefloquine, amodiaquine, artemisinin, metronidazole, ampicillin
Contraindications
- Known or suspected chloroquine resistance in P. falciparum
- Hypersensitivity
- Retinal damage
- Concurrent therapy with hepatotoxic drugs
Side Effects
- Gastro-intestinal symptoms: Nausea, vomiting, diarrhoea
- Psychological symptoms: Psychotic episodes, anxiety, personality changes
- Visual disturbances: Blurred vision, difficulties in focusing, retinopathy
- Haematological symptoms: Aplastic anaemia, agranulocytosis, thrombocytopenia
- Other symptoms: Loss of hair, pigmentation of the mucous membranes and skin, photosensitivity, tinnitus, reduced hearing, nerve deafness
Pregnancy & Lactation
- Avoid use during pregnancy except in the treatment of malaria if benefits outweigh hazards
- Consideration needed for discontinuation of nursing if taking chloroquine
Precautions & Warnings
- Examination of eyes before starting long-term treatment
- Monitoring of visual disturbances
- Care with impaired liver or renal function, porphyria, psoriasis, or history of epilepsy
- Observation for haemolytic anaemia in patients with G6PD deficiency during chloroquine treatment
