Indications
- Treatment of malaria

Composition
- Film coated tablets with Chloroquine Phosphate BP 250 mg
- Syrup with Chloroquine Phosphate BP 80 mg
Mode of Action
- 4-aminoquinoline antimalarial and amoebicidal agent
- Suppresses and clinically cures malaria due to susceptible strains of Plasmodium
- Rapidly acting blood schizontocide with gametocytocidal activity
- Raises intravesicular pH in malaria parasite cells
- Interferes with synthesis of nucleoproteins by the parasite
- Rapid and complete absorption from the gastrointestinal tract
Dosage & Administration
- Doses expressed in terms of chloroquine base
- Oral dose equivalent to 300 mg chloroquine base for clinical prophylaxis
- Weekly oral dose of 5 mg per kg body-weight for children in prophylaxis
- 600 mg base daily for two days, followed by 300 mg base daily for at least 2-3 weeks for hepatic amoebiasis
- 150 mg of base daily for discoid and systemic lupus erythematosus
- 150 mg of base daily for rheumatoid arthritis
- 150 to 300 mg of base daily for the management of photoallergic reactions
- 75 mg of base 2 to 3 times per week for 6 to 18 months for porphyria cutanea tarda
Interaction
- Interacts with neostigmine, pyridostigmine, antacids, kaolin, cimetidine, ranitidine, quinine, mefloquine, amodiaquine, artemisinin, metronidazole, ampicillin
Contraindications
- Known or suspected chloroquine resistance in P. falciparum
- Hypersensitivity
- Retinal damage
- Concurrent therapy with hepatotoxic drugs
Side Effects
- Nausea, vomiting, and diarrhea
- Psychotic episodes, anxiety, and personality changes
- Blurred vision and difficulties in focusing
- Aplastic anemia, agranulocytosis, thrombocytopenia, and neutropenia
- Loss of hair, bleaching of hair pigment, pigmentation of skin, photosensitivity, tinnitus, and reduced hearing
Pregnancy & Lactation
- Avoid use during pregnancy except for malaria suppression or treatment
- Potential for serious adverse reactions in nursing infants
Precautions & Warnings
- Eyes should be examined before long-term treatment
- Visual disturbances may affect ability to operate vehicles or machinery
- Care needed for patients with impaired liver or renal function, prophyria, psoriasis, or a history of epilepsy
- Observation for haemolytic anaemia in patients with G6PD deficiency
