Indications
- Uncomplicated, severe and complicated falciparum malaria
- Falciparum malaria resistant to other drugs
- Prevention of nocturnal muscle cramps
- Alleviation of muscle cramps in Thomsen's disease
Pharmacology
Interferes with protein metabolism, inhibits nucleic acid synthesis in malarial parasites, highly active blood schizonticide
Dosage & Administration
- Adult:
- Oral: 600 mg every 8 hours for 7 days
- Parenteral: Loading dose of 20 mg/kg (up to max 1.4 g) infused over 4 hours, maintenance dose of 10 mg/kg (up to max 700 mg) infused over 4 hours every 8-12 hours
- Children:
- Oral: 10 mg/kg every 8 hours for 7 days
- Parenteral: Calculated on a mg/kg basis as for adult
- Pregnancy: Treatment doses given to pregnant women
Contraindications
Hypersensitivity reaction, optic neuritis, black water fever, etc.
Side Effects
Cinchonism (tinnitus, headache, nausea, visual disturbances), hematological disorders, skin reaction
Pregnancy & Lactation
Considered safe when used in therapeutic dosage in pregnancy, excreted in breast milk in insignificant amount
Precautions & Warnings
- Severe heart disease, myasthenia gravis, diabetes mellitus
- IV injection should be given slowly
- Use of Mefloquine with Jasoquin sulphate may increase the chance of side effects
