Indications
- Prophylaxis against arterial occlusive events
- Mild to moderate pain
- Chronic disease accompanied by pain and inflammation
- Antipyretic
Pharmacology
- Decreases platelet aggregation
- Inhibits thrombus formation on the arterial side of the circulation
- Analgesic for headache and musculoskeletal pain
- Anti-inflammatory
- Antipyretic
- Enteric-coated to reduce intestinal disturbance and gastrointestinal ulceration
Dosage & Administration
- Aspirin 300 mg 1-3 tablets 6 hourly with a maximum daily dose of 4 gm for pain, inflammatory diseases, and as antipyretic
- 150 mg-300 mg immediately for suspected acute coronary syndrome
- 150 mg daily for 1 month after myocardial infarction
- 75 mg daily for long-term use after myocardial infarction
- 150 mg-300 mg daily for acute ischaemic stroke/transient ischaemic stroke (TIA)
- 75 mg-300 mg daily starting 6 hours post-bypass surgery
Interaction
- Enhances effect of anticoagulants, oral hypoglycaemic agents, phenytoin, and sodium valproate
- Inhibits uricosuric effect of probenecid
- May increase the toxicity of sulphonamides
- May precipitate bronchospasm or induce asthma attacks
Contraindications
- Children less than 12 years (Reye's syndrome)
- Breastfeeding
- Active peptic ulcer
- Bleeding due to haemophilia
- Intracranial haemorrhage
- Other ulceration
Side Effects
- Mild side effects including nausea, dyspepsia, gastrointestinal ulceration, and bronchospasm
Pregnancy & Lactation
- Avoid use during the last 3 months of pregnancy
- Caution in lactating mothers
Precautions & Warnings
- Administer cautiously in asthma, uncontrolled blood pressure, and pregnant women
- Administer with caution to patients with nasal polyp and nasal allergy
Overdose Effects
- Dizziness
- Tinnitus
- Sweating
- Nausea and vomiting
- Confusion
- Hyperventilation
- CNS depression with coma
- Cardiovascular collapse
- Respiratory depression
- Treatment includes gastric lavage and forced alkaline diuresis, haemodialysis in severe cases
