Indications
- Carcinoma of the colon or rectum

Pharmacology
Fluorouracil is converted into the active 5-fluorodeoxyuridine monophosphate (FdUMP) by a variety of different metabolic pathways. It inhibits the enzyme thymidylate kinase, reduces DNA formation, and is incorporated into RNA resulting in fluorination of RNA.
Dosage & Administration
- Dosage of 12-13.5 mg/Kg (500 mg/m2) daily for 5 days repeated at 4-weekly intervals
- Intravenous administration over 2-3 hr on 5 successive days
- Intra-arterial infusion options
- Combination with radiation or other anticancer drugs
Interaction
Pre-treatment with cimetidine may increase plasma concentrations of 5-Fluril.
Contraindications
Contraindicated in severely debilitated patients, those with bone marrow suppression, pregnant or breastfeeding women, and non-malignant illnesses.

Pregnancy & Lactation
Contraindicated throughout pregnancy and breastfeeding due to risk of teratogenesis and inhibition of DNA, RNA, and protein synthesis.
Precautions & Warnings
5-Fluril is highly toxic and therapy should be discontinued promptly when signs of toxicity appear.
Use in Special Populations
- No dosage recommendations for neonates
- Safety and effectiveness not established in children
- Doses adjusted for the elderly
Overdose Effects
No specific antidote to 5-Fluril toxicity; treatment consists of supportive care.
Reconstitution
Special personnel and precautions required for reconstitution; handling should be done with protective equipment and careful disposal of contaminated waste.
Storage Conditions
Vial should be stored in original carton at temperatures not exceeding 25℃, protected from light.
