Pharmacology

• Combination of Paracetamol and Caffeine
• Analgesic and antipyretic properties of Paracetamol
• Increased solubility and transmembrane permeation of Paracetamol due to intermolecular association with Caffeine
• Increase in pain threshold and tolerance of pain
• Beneficial for treating migraine and headache with its power to raise vessel tone in the brain

DosageAdministration

Adult dose: 1-2 tablets every 4-6 hours. Maximum dose: 8 tablets daily. Child dose: Not recommended for children below 12 years.

Interaction

• May reduce serum levels with anticonvulsants
• Enhanced anticoagulant effect with prolonged use of warfarin and other coumarins
• Accelerated absorption with metoclopramide and domperidone
• Increased serum levels with probenecid
• Increased serum levels of chloramphenicol
• Reduced absorption with colestyramine within 1 hr of admin
• Potential to cause severe hypothermia with phenothiazine

Contraindications

• Severe renal function impairment
• Hepatic disease (Viral Hepatitis)
• Known hypersensitivity to paracetamol or caffeine

SideEffects

• Mild side effects of paracetamol
• Haematological reactions including thrombocytopenia, leukopenia, pancytopenia, neutropenia, and agranulocytosis
• Occasional occurrences of pancreatitis, skin rashes, and other allergic reactions

PregnancyLactation

• Consultation required for pregnant mothers
• Can be taken while breastfeeding

PrecautionsWarnings

• Cautious administration in patients with hepatic or renal failure
• Cautious use in patients taking other hepatotoxic medication
• Avoid prolonged use without consulting a physician

OverdoseEffects

• Symptoms of overdose in the first 24 hours: pallor, nausea, vomiting, anorexia, and abdominal pain
• Liver damage may appear 12 to 40 hours after ingestion
• Abnormalities of glucose metabolism and metabolic acidosis
