Pharmacology
- Combination of Paracetamol and Caffeine
- Analgesic and antipyretic properties
- Weak anti-inflammatory activity
- Caffeine increases solubility and transmembrane permeation of Paracetamol
- Increases pain threshold and tolerance of pain
- Raises vessel tone in the brain
- Beneficial for treating migraine and headache
Dosage & Administration
- Adult dose: 1-2 tablets every 4-6 hours
- Maximum daily dose: 8 tablets
- Not recommended for children below 12 years
Interaction
- May reduce serum levels with anticonvulsants
- May enhance anticoagulant effect of warfarin and other coumarins
- Accelerated absorption with metoclopramide and domperidone
- May increase serum levels with probenecid
- May increase serum levels of chloramphenicol
- May reduce absorption with colestyramine within 1 hr of admin
- May cause severe hypothermia with phenothiazine
Contraindications
- Contraindicated in severe renal function impairment
- Contraindicated in hepatic disease (Viral Hepatitis)
- Known hypersensitivity to paracetamol or caffeine
Side Effects
- Mild side effects of paracetamol
- Haematological reactions including thrombocytopenia, leukopenia, pancytopenia, neutropenia, and agranulocytosis
- Pancreatitis, skin rashes, and allergic reactions occur occasionally
Pregnancy & Lactation
- Consultation required for pregnant mothers
- Safe for breastfeeding mothers
Precautions & Warnings
- Cautious use in patients with hepatic or renal failure
- Cautious use in patients taking other hepatotoxic medication
- Avoid prolonged use without consulting a physician
Overdose Effects
- Symptoms include pallor, nausea, vomiting, anorexia, and abdominal pain
- Liver damage may occur 12 to 40 hours after ingestion
- Abnormalities of glucose metabolism and metabolic acidosis may occur
