Pharmacology

Centrally acting synthetic analgesic compound

Mode of Action

Inhibits reuptake of neurotransmitters - serotonin and noradrenaline
Modifies transmission of pain impulses
Stimulates mu-opioid receptors
Modulates central monoaminergic inhibitory pathways

Dosage & Administration

Usual doses of 50 to 100 mg every 4 to 6 hours
Initial dose of 100 mg for acute pain, 50 mg for chronic pain
Sustained Release Capsule: One SR capsule every 12 hours
Injection dose of 50-100 mg every 4 to 6 hours
Suppository for adults: usual dose of 100 mg every 6 hours

Interaction

Contraindicated with monoamine oxidase (MAO) inhibitors

Hypersensitivity to the drug
Acute intoxication with alcohol, hypnotics, opioids, or psychotropic drugs
