Indications
- Treatment of moderate to severe painful conditions
- Postoperative pain
- Colic and spastic pain
- Cancer pain
- Joint pain
- Neck and back pain
- Pain associated with osteoporosis
Pharmacology
- Centrally acting synthetic analgesic compound
- Inhibits reuptake of serotonin and noradrenaline
- Modifies transmission of pain impulses
- Activates descending serotonergic pathways
- Stimulation of mu-opioid receptors
- Modulation of central monoaminergic pathways
Dosage & Administration
- Usual doses of 50 to 100 mg every 4-6 hours
- Initial dose of 100 mg for acute pain
- Initial dose of 50 mg for chronic pain
- Sustained Release Capsule or Tablet for long-lasting relief
- Injection for rapid pain relief
- Suppository for rectal administration
Interaction
- Monoamine oxidase (MAO) inhibitors not recommended for combination
- Carbamazepine interaction requires dose adjustment
Contraindications
- Hypersensitivity to the drug
- Acute intoxication with certain substances

Precautions & Warnings
- Cautious use in patients at risk for respiratory depression
- Not recommended for opioid-dependent patients
- Caution with concomitant CNS depressants
- Great caution with concomitant MAO inhibitors
- Use with caution in patients with increased intracranial pressure or head injury
Use in Special Populations
- Child dosage of 1-2 mg/kg body weight
- Suppository not for children below 14 years
- Safety and efficacy not established in children for certain products
