Indications
- Postoperative pain
- Colic and spastic pain

Pharmacology
- Centrally acting synthetic analgesic
- Inhibits reuptake of neurotransmitters (serotonin and noradrenaline)
- Modifies transmission of pain impulses
- Stimulation of mu-opioid receptors
- Activation of serotonergic and noradrenergic pathways
Dosage & Administration
- Dosage forms: Capsule, tablet, sustained release capsule/tablet, injection, suppository
- Doses of 50-100 mg every 4-6 hours
- Initial dose of 100 mg for acute pain, 50 mg for chronic painful conditions
- Usual dosing frequency
- Max daily dosage by mouth: 400 mg
- Dosing adjustment for postoperative pain
Interaction
- Interaction with monoamine oxidase (MAO) inhibitors
- Increased metabolism with carbamazepine
Contraindications
- Hypersensitivity to tramadol
- Contraindicated in acute intoxication with certain substances

Precautions & Warnings
- Respiratory depression risk
- Risk in patients dependent on opioids
- Caution with concomitant CNS depressants
- Caution with concomitant MAO inhibitors
- Caution in patients with increased intracranial pressure or head injury
- Caution in patients with acute abdominal conditions
Use in Special Populations
- Dosage for children over 1 year old
- Suppository not recommended for children under 14 years old
- Safety and efficacy not established in children for specific product
