Indications
- Management of moderate to moderately severe pain in adults
- Short-term management of acute pain
Pharmacology
- Paracetamol with analgesic and antipyretic properties
- Weak anti-inflammatory activity
- Thought to act primarily in the CNS
- Increases pain threshold by inhibiting cyclooxygenase (COX-1, COX-2, COX-3) enzymes
- Tramadol - centrally acting synthetic opioid analgesic
- Mode of action not completely understood
- Binding to μ-opioid receptors
- Inhibition of norepinephrine and serotonin reuptake
Dosage & Administration
- Recommended dose for moderate to moderately severe pain
- Recommended dose for short-term (five days or less) management of acute pain
- Can be administered without regard to food
Contraindications
- Hypersensitivity to tramadol, paracetamol, or opioids
- Contraindicated in situations where opioids are contraindicated

Precautions & Warnings
- May impair mental or physical abilities
- Should not be taken with alcohol
- Should not be taken in combination with other tramadol or paracetamol-containing products
- Use with caution when taking tranquilizers, hypnotics or other opiate containing analgesics
Use in Special Populations
- Safety and effectiveness not studied in the pediatric population
- Dose selection for elderly patients should be cautious
- Not studied in patients with impaired renal function
- In patients with creatinine clearances of less than 30 ml/min, dosing interval should be increased
- Not studied in patients with impaired hepatic function
- Not recommended for use in patients with hepatic impairment
