Pharmacology
- Inhibits peripheral actions of serotonin and histamine
- Increases membrane permeability of cranial vessels
- Alters pain thresholds in migraines
- Blocks 5-HT receptors
- Attenuates signaling of serotonin in cranial vasoconstriction
- Inhibits peripheral actions of bradykinin
- May inhibit serotonin reuptake by blood platelets
- Decreases passive distension of extracranial arteries
- Appetite stimulation at the metabolic level
Dosage & Administration
- Adults: Usually 1.5 mg daily
- May be taken as a single dose at night or in three divided doses
- Maximum of 4.5 mg daily for adults
- Up to 3 mg may be given as a single dose for adults
- Children (aged over 2 years): Up to 1.5 mg daily
- Usually as a divided dose for children
- Up to 1 mg has been given as a single dose at night for children
Interaction
- Enhanced central effects of sedatives, hypnotics, antihistamines, and alcohol
- Antagonizes hypotensive effect of adrenergic neurone blockers
Contraindications
- Known hypersensitivity to any of its ingredients
