Description

  • Tricyclic compound
  • Antagonizes serotonin receptor competitively
  • Antihistamine properties
  • Antibradykinin properties
  • Weak anticholinergic properties
  • Appetite-stimulating properties

Pharmacology

  • Inhibits peripheral actions of serotonin and histamine
  • Increases membrane permeability of cranial vessels
  • Alters pain thresholds in migraines
  • Blocks 5-HT receptors
  • Attenuates signaling of serotonin
  • Inhibits serotonin reuptake by blood platelets
  • Affects tonicity and decreases passive distension of extracranial arteries
  • Inhibits peripheral actions of bradykinin
  • May stimulate appetite at metabolic level

Dosage & Administration

  • Adults: Usually 1.5 mg daily
  • May be taken as a single dose at night or in three divided doses
  • Dosage should be adjusted to individual patient requirements
  • Maximum dosage of 4.5 mg daily
  • Children (aged over 2 years): Up to 1.5 mg daily
  • Usually as a divided dose
  • Up to 1 mg given as a single dose at night

Interaction

  • Enhances central effects of sedatives, hypnotics, antihistamines
  • Enhances effects of alcohol
  • Antagonizes the hypotensive effect of adrenergic neurone blockers

Contraindications

  • Known hypersensitivity to any of its ingredients
