Description
- Tricyclic compound
- Antagonizes serotonin receptor competitively
- Antihistamine properties
- Antibradykinin properties
- Weak anticholinergic properties
- Appetite-stimulating properties
Pharmacology
- Inhibits peripheral actions of serotonin and histamine
- Increases membrane permeability of cranial vessels
- Alters pain thresholds in migraines
- Blocks 5-HT receptors
- Attenuates signaling of serotonin in causing cranial vasoconstriction
- Inhibits peripheral actions of bradykinin
- May inhibit serotonin reuptake by blood platelets
- Affects tonicity and decreases passive distension of extracranial arteries
- Leads to appetite stimulation
Dosage & Administration
- Adult dosage: Usually 1.5 mg daily
- May be taken as a single dose at night or in three divided doses
- Dosage should be adjusted to individual patient requirements
- Maximum dosage of 4.5 mg daily
- Children's dosage: Up to 1.5 mg daily
Interaction
- Enhances central effects of sedatives, hypnotics, antihistamines, and alcohol
- Antagonizes hypotensive effect of adrenergic neurone blockers

- Hypersensitivity to any of its ingredients
