- Antagonizes serotonin receptor competitively

Pharmacology
- Inhibits peripheral actions of serotonin and histamine
- Increases membrane permeability of cranial vessels
- Alters pain thresholds in migraines
- Blocks 5-HT receptors
- Attenuates serotonin signaling in causing cranial vasoconstriction
- Inhibits peripheral actions of bradykinin
- May inhibit serotonin reuptake by blood platelets
- Decreases passive distension of extracranial arteries
- Affects tonicity of extracranial arteries
- Appetite stimulation at metabolic level
Dosage & Administration
- Adult dosage: usually 1.5 mg daily
- May be taken as a single dose at night or in three divided doses
- Maximum of 4.5 mg daily
- Up to 3 mg may be given as a single dose
- Children dosage: up to 1.5 mg daily
- Usually as a divided dose
- Up to 1 mg has been given as a single dose at night
Interaction
- Enhances central effects of sedatives and hypnotics
- Enhances effects of antihistamines and alcohol
- Antagonizes the hypotensive effect of adrenergic neurone blockers
