Pharmacology

  • Inhibits peripheral actions of serotonin and histamine
  • Alters pain thresholds in migraines
  • Blocks 5-HT receptors
  • Attenuates serotonin signaling for cranial vasoconstriction
  • Inhibits peripheral actions of bradykinin
  • May inhibit serotonin reuptake by blood platelets
  • Affects tonicity and decreases passive distension of extracranial arteries
  • Appetite-stimulating effects at metabolic level

Dosage & Administration

  • Adult dose: Usually 1.5 mg daily
  • Maximum adult dose: 4.5 mg daily
  • Children aged over 2 years: Up to 1.5 mg daily
  • Dosage adjustment based on individual patient requirements
  • May be taken as a single dose at night or in divided doses
  • Up to 3 mg may be given as a single dose

Interaction

  • Enhanced effects with sedatives, hypnotics, antihistamines, and alcohol
  • Antagonizes the hypotensive effect of adrenergic neurone blockers
