Description

  • Tricyclic compound
  • Antagonizes serotonin receptor
  • Antihistamine properties
  • Antibradykinin properties
  • Weak anticholinergic properties
  • Appetite-stimulating properties

Pharmacology

  • Inhibits peripheral actions of serotonin and histamine
  • Increases membrane permeability of cranial vessels
  • Alters pain thresholds in migraines
  • Blocks 5-HT receptors
  • Attenuates signalling of serotonin in causing cranial vasoconstriction
  • Inhibits peripheral actions of bradykinin
  • May inhibit serotonin reuptake by blood platelets
  • Decreases passive distension of extracranial arteries

Dosage & Administration

  • Adult dosage: Usually 1.5 mg daily
  • May be taken as a single dose at night or in three divided doses
  • Maximum dosage of 4.5 mg daily
  • Up to 3 mg may be given as a single dose
  • Children dosage: Up to 1.5 mg daily (divided doses)

Interaction

  • Central effects of sedatives, hypnotics, antihistamines, and alcohol may be enhanced
  • Antagonizes the hypotensive effect of adrenergic neurone blockers

Contraindications

  • Known hypersensitivity to any of its ingredients
