Indications
- Prophylactic treatment of vascular headache of migraine type such as classical migraine, common migraine, and cluster headache
Description
- Tricyclic compound
- Antagonizes serotonin receptor competitively
- Possesses antihistamine, antibradykinin, and weak anticholinergic properties
- Appetite-stimulating properties
Pharmacology
- Inhibits peripheral actions of serotonin and histamine in increasing the membrane permeability of cranial vessels and transudation of plasmakinin
- Alters pain thresholds in migraines
- Blocks 5-HT receptors to attenuate the signaling of serotonin causing cranial vasoconstriction
- Inhibits peripheral actions of bradykinin
- May inhibit serotonin reuptake by blood platelets
- Affects tonicity and decreases passive distension of extracranial arteries
- May stimulate appetite by acting at the metabolic level rather than direct stimulation of the appetite center
Dosage & Administration
- Adults: Usually 1.5 mg daily, may be taken as a single dose at night or in three divided doses, with a maximum of 4.5 mg daily and up to 3 mg as a single dose
- Children (aged over 2 years): Up to 1.5 mg daily, usually as a divided dose or up to 1 mg as a single dose at night
Interaction
- Enhances central effects of sedatives, hypnotics, antihistamines, and alcohol
- Antagonizes the hypotensive effect of adrenergic neurone blockers
Contraindications
- Known hypersensitivity to any of its ingredients
