Description
- Tricyclic compound
- Antagonizes serotonin receptor competitively
- Antihistamine properties

Pharmacology
- Inhibits peripheral actions of serotonin and histamine
- Increases membrane permeability of cranial vessels
- Alters pain thresholds in migraines
- Attenuates serotonin signaling causing cranial vasoconstriction
- Inhibits serotonin reuptake by blood platelets
- Affects tonicity and decreases passive distension of extracranial arteries
- Inhibits peripheral actions of bradykinin
- May act at the metabolic level to stimulate appetite
Dosage & Administration
- Adult dosage: 1.5 mg daily
- Can be taken as a single dose at night or in three divided doses
- Maximum dosage: 4.5 mg daily
- Children dosage: Up to 1.5 mg daily (aged over 2 years)
- Higher dosage for children may be given as a single dose at night
Interaction
- Enhances central effects of sedatives, hypnotics, and antihistamines
- Antagonizes the hypotensive effect of adrenergic neurone blockers
Contraindications
- Contraindicated in patients with known hypersensitivity to any of its ingredients
