Description
- Tricyclic compound
- Antagonizes serotonin receptor
- Antihistamine properties
- Antibradykinin properties

Pharmacology
- Inhibits peripheral actions of serotonin and histamine
- Increases membrane permeability of cranial vessels
- Alters pain thresholds in migraines
- Blocks 5-HT receptors
- Attenuates signaling of serotonin
- Inhibits peripheral actions of bradykinin
- May inhibit serotonin reuptake by blood platelets
- Decreases passive distension of extracranial arteries
- Stimulates appetite at the metabolic level
Dosage & Administration
- Adult dosage: 1.5 mg daily
- Can be taken as a single dose at night or in three divided doses
- Maximum dosage of 4.5 mg daily
- 3 mg maximum single dose
- Children dosage: Up to 1.5 mg daily
- Usually as a divided dose for children
- Up to 1 mg as a single dose at night for children
Interaction
- Enhances central effects of sedatives, hypnotics, antihistamines, and alcohol
- Antagonizes the hypotensive effect of adrenergic neurone blockers
Contraindications
- Contraindicated in patients with known hypersensitivity to any of its ingredients

Pregnancy & Lactation
- Limited information on using Pizotifen in pregnancy
- No reported ill effects in pregnancy
- Given during pregnancy only if potential benefit justifies potential risk to fetus
