Indications
- Symptomatic relief of Osteoarthritis (OA)
- Symptomatic relief of Rheumatoid arthritis (RA)
- Symptomatic relief of Ankylosing spondylitis
- Symptomatic relief of acute gouty arthritis
- Short-term treatment of moderate pain associated with dental surgery
Pharmacology
- Potent orally active cyclooxygenase-2 (COX-2) specific inhibitor
- Inhibition of COX-2 decreases signs and symptoms of pain, inflammation, and fever
- Decreased potential for gastrointestinal toxicity and effects on platelet aggregation
Dosage & Administration
- Recommended dose for Osteoarthritis: 30 mg once daily, may increase to 60 mg once daily if insufficient relief
- Recommended dose for Rheumatoid arthritis and Ankylosing spondylitis: 90 mg once daily
- Recommended dose for Acute gouty arthritis: 120 mg once daily for 8 days
- Recommended dose for Postoperative dental surgery pain: 90 mg once daily up to 3 days
Interaction
- Increase in prothrombin time with oral anticoagulants
- Reduction in effect of diuretics and other antihypertensive drugs with NSAIDs
- Concomitant use with low-dose acetylsalicylic acid
- Potential increase in nephrotoxic effect when coadministered with ciclosporin or tacrolimus
- Decreased lithium renal excretion and increased lithium plasma levels with NSAIDs
- Take without regards to meals
Contraindications
- Hypersensitivity to the active substance or excipients
- Active peptic ulceration or gastrointestinal (GI) bleeding
- History of allergic-type reactions to acetylsalicylic acid or NSAIDs
- Pregnancy, lactation, severe hepatic dysfunction, estimated renal creatinine clearance <30 ml/min
- Children and adolescents under 16 years of age
- Inflammatory bowel disease, congestive heart failure, uncontrolled hypertension
- Established ischaemic heart disease, peripheral arterial disease, cerebrovascular disease

Precautions & Warnings
- Caution in patients at risk of developing gastrointestinal complications
- Special consideration for patients with significant cardiovascular risk factors
- Monitoring of renal function in patients with decreased prostaglandin formation
- Caution in patients with history of cardiac failure, left ventricular dysfunction, hypertension, or edema
- Monitoring of liver function and discontinuation upon signs of hepatic insufficiency
- Discontinuation at first appearance of hypersensitivity signs
- Caution when co-administering with warfarin or other oral anticoagulants
Overdose Effects
- Single doses up to 500 mg and multiple doses up to 150 mg/day did not result in significant toxicity
- Usual supportive measures may be employed in the event of overdose
