Indications
- Reduction of total cholesterol and LDL cholesterol
- Reduction of apolipoprotein B (Apo-B) and triglycerides levels
- Treatment of familial hypercholesterolaemia
- Treatment of mixed dyslipidemia (Fredrickson Type Ia and Ib)
- Treatment of hypertriglyceridaemia (Fredrickson Type IV)
- Treatment of dysbetalipoproteinaemia (Fredrickson Type III)
- Reduction of cardiac ischaemic events in patients with coronary artery disease
- Reduction of total and LDL-cholesterol concentrations in patients with hypercholesterolemia associated with diabetes mellitus or renal transplantation
Pharmacology
- Selective inhibitor of HMG-CoA reductase
- Lowers plasma cholesterol and lipoprotein levels
- Increases hepatic LDL receptors for enhanced uptake of LDL
- Rapid absorption after oral administration
Dosage & Administration
- 10 mg once daily for adults
- 10-80 mg once daily for adults with necessary dosage adjustments
- 10-20 mg once daily for children (10-18 years)
- Dosage range of 10-80 mg once daily
- Administration as a single dose at any time of the day, with or without food
- Regular lipid level analysis within 2 to 4 weeks after initiation
Interactions
- Increased risk of myopathy with concurrent administration of cyclosporine, fibric acid derivatives, niacin, erythromycin, azole antifungals
- Altered plasma concentrations when coadministered with antacids, colestipol, digoxin, erythromycin, oral contraceptives, warfarin
Contraindications
- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors

Precautions & Warnings
- Liver function tests should be performed before initiation and periodically thereafter
- Use with caution in patients with substantial alcohol consumption or history of liver disease
- Discontinue therapy if markedly elevated CPK levels occur or myopathy is diagnosed
Use in Special Populations
- Higher plasma concentrations in healthy elderly subjects
- Greater LDL-lowering in the elderly patient population
- No influence of renal disease on plasma concentrations or LDL-C reduction
- No significant enhancement of clearance with hemodialysis
