Indications
- Symptomatic relief of osteoarthritis
- Symptomatic relief of rheumatoid arthritis
- Symptomatic relief of ankylosing spondylitis
- Symptomatic relief of acute gouty arthritis
- Short-term treatment of moderate pain associated with dental surgery
Pharmacology
- Potent COX-2 specific inhibitor
- Targets prostaglandin-mediated pain, inflammation, and fever
- Decreases signs and symptoms with decreased potential for GI toxicity and effects on platelet aggregation
Dosage & Administration
- Recommended doses for osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, acute gouty arthritis, and postoperative dental surgery pain
- Additional postoperative analgesia may be required
- Shortest duration and lowest effective daily dose should be used
Interaction
- Interaction with oral anticoagulants
- Interaction with diuretics, ACE inhibitors, and Angiotensin II Antagonists
- Concomitant use with acetylsalicylic acid for cardiovascular prophylaxis
- Potential interaction with ciclosporin, tacrolimus, and lithium
Contraindications
- Hypersensitivity to the active substance or excipients
- Active peptic ulceration or gastrointestinal bleeding
- History of allergic-type reactions to aspirin or NSAIDs
- Pregnancy and lactation
- Severe hepatic dysfunction
- Estimated renal creatinine clearance <30 ml/min
- Inflammatory bowel disease
- Congestive heart failure
- Uncontrolled hypertension
- Established ischemic heart disease, peripheral arterial disease, and/or cerebrovascular disease

- Caution in patients with history of cardiac failure, left ventricular dysfunction, or hypertension
- Monitoring advised for signs of hepatic insufficiency
- Discontinue at first appearance of hypersensitivity
- Potential masking of fever and signs of inflammation
Overdose Effects
- No significant toxicity observed at single doses up to 500 mg and multiple doses up to 150 mg/day for 21 days
- Usual supportive measures advised in case of overdose
