Indications
- Symptomatic relief of osteoarthritis
- Symptomatic relief of rheumatoid arthritis
- Symptomatic relief of ankylosing spondylitis
- Symptomatic relief of acute gouty arthritis
- Short-term treatment of moderate pain associated with dental surgery
Pharmacology
- Potent orally active COX-2 specific inhibitor
- Inhibition of COX-2 decreases clinical signs and symptoms
- Selective inhibition of COX-2 with decreased potential for GI toxicity
- Did not inhibit gastric prostaglandin synthesis
Dosage & Administration
- Recommended doses for osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, acute gouty arthritis, and postoperative dental surgery pain
- Dose adjustments for insufficient relief from symptoms
- Shortest duration possible and lowest effective daily dose recommended
Interactions
- Interaction with oral anticoagulants
- Interaction with diuretics, ACE inhibitors, and Angiotensin II antagonists
- Concomitant use with acetylsalicylic acid for cardiovascular prophylaxis
- Potential interaction with ciclosporin, tacrolimus, and lithium
Contraindications
- Hypersensitivity to active substance or excipients
- Active peptic ulceration or active gastrointestinal bleeding
- History of allergic-type reactions to acetylsalicylic acid or NSAIDs
- Pregnancy, lactation, severe hepatic dysfunction, and estimated renal creatinine clearance <30 ml/min
- Inflammatory bowel disease, congestive heart failure, uncontrolled hypertension, and established cardiovascular diseases
Side Effects
- Palpitation, fatigue, influenza-like symptoms, ecchymosis
- Less commonly dry mouth, taste disturbance, mouth ulcer, atrial fibrillation
- Very rarely confusion and hallucinations
Pregnancy & Lactation
- Not recommended in women attempting to conceive
- Excreted in the milk of lactating rats, women who use Etoricoxib must not breastfeed
Precautions & Warnings
- Caution with patients at risk of gastrointestinal complications
- Consideration for patients with significant cardiovascular risk factors
- Monitoring of renal function in patients and caution in patients with cardiac failure, hypertension, and edema
- Monitoring of liver function, discontinuation at signs of hypersensitivity, caution with warfarin co-administration
Overdose Effects
- No significant toxicity observed with single and multiple doses administered
- Employment of usual supportive measures in case of overdose
