Indications
- Symptomatic relief of Osteoarthritis (OA)
- Rheumatoid arthritis (RA)
- Ankylosing spondylitis
- Pain and signs of inflammation associated with acute gouty arthritis
- Short-term treatment of moderate pain associated with dental surgery
Pharmacology
- Potent, orally active cyclooxygenase-2 (COX-2) specific inhibitor
- Selective inhibition of COX-2 within the clinical dose range
- Decreases clinical signs and symptoms with decreased potential for GI toxicity and effects on platelet aggregation
- Did not inhibit gastric prostaglandin synthesis
Dosage & Administration
- Recommended doses for different conditions: 30mg once daily, 60mg once daily, 90mg once daily, 120mg once daily
- Shortest duration possible and lowest effective daily dose should be used
- Re-evaluate periodically, especially in patients with osteoarthritis
Interaction
- Oral anticoagulants: associated with an increase in prothrombin time
- NSAIDs may reduce the effect of diuretics and other antihypertensive drugs
- Can be used concomitantly with low-dose acetylsalicylic acid for cardiovascular prophylaxis
- Coadministration with ciclosporin or tacrolimus may increase nephrotoxic effect
- Decreases lithium renal excretion and increases lithium plasma levels
Contraindications
- Hypersensitivity to the active substance or to any of the excipients
- Active peptic ulceration or active gastrointestinal (GI) bleeding
- Patients with certain allergic-type reactions after taking NSAIDs
- Pregnancy and lactation
- Severe hepatic dysfunction
- Estimated renal creatinine clearance <30 ml/min
- Children and adolescents under 16 years of age
- Inflammatory bowel disease
- Congestive heart failure, hypertension, established cardiovascular diseases

Pregnancy & Lactation
- Not recommended in women attempting to conceive
- Excreted in the milk of lactating rats
- Women who use Etoricoxib must not breastfeed
Precautions & Warnings
- Caution in patients at risk of developing a gastrointestinal complication with NSAIDs
- Treatment in patients with significant risk factors for cardiovascular events after careful consideration
- May cause reduction in prostaglandin formation and impair renal function
- Caution in patients with history of cardiac failure, left ventricular dysfunction, or hypertension
- Monitoring of liver function and discontinuation if signs of hepatic insufficiency occur
Overdose Effects
- Single doses up to 500 mg and multiple doses up to 150 mg/day did not result in significant toxicity
- Usual supportive measures in the event of overdose: remove unabsorbed material, employ clinical monitoring, institute supportive therapy
