Indications
- Symptomatic relief of osteoarthritis (OA)
- Symptomatic relief of rheumatoid arthritis (RA)
- Symptomatic relief of ankylosing spondylitis
- Symptomatic relief of acute gouty arthritis
- Short-term treatment of moderate pain associated with dental surgery
Pharmacology
- Etoricoxib is a potent orally active COX-2 inhibitor
- Inhibition of COX-2 reduces clinical signs and symptoms
- Selective inhibition of COX-2 decreases potential for GI toxicity
- Etoricoxib does not inhibit gastric prostaglandin synthesis
Dosage & Administration
- Recommended dose for osteoarthritis: 30mg once daily
- Recommended dose for rheumatoid arthritis: 90mg once daily
- Recommended dose for ankylosing spondylitis: 90mg once daily
- Recommended dose for acute gouty arthritis: 120mg once daily
- Recommended dose for postoperative dental surgery pain: 90mg once daily, max 3 days
- Shortest duration possible and lowest effective daily dose should be used
Interactions
- Interaction with oral anticoagulants
- Interaction with diuretics, ACE inhibitors, and Angiotensin II Antagonists
- Can be used concomitantly with low-dose acetylsalicylic acid
- Potential interaction with ciclosporin and tacrolimus
- NSAIDs may decrease lithium renal excretion
Contraindications
- Hypersensitivity to the active substance or excipients
- Active peptic ulceration or active GI bleeding
- History of allergic-type reactions to NSAIDs
- Pregnancy and lactation
- Severe hepatic dysfunction
- Estimated renal creatinine clearance <30 ml/min
- Children and adolescents under 16 years of age
- Inflammatory bowel disease
- Congestive heart failure
- Hypertension, ischaemic heart disease, peripheral arterial disease, and/or cerebrovascular disease

- Not recommended for women attempting to conceive

Precautions & Warnings
- Caution with patients at risk of gastrointestinal complications
- Caution with patients at significant risk of cardiovascular events
- May impair renal function and require monitoring
- Caution with patients with history of cardiac failure, left ventricular dysfunction, or hypertension
Overdose Effects
- Single doses of up to 500mg and multiple doses up to 150mg/day did not result in significant toxicity
