Description
Astin 10 mg Tablet is used as an adjunct to diet to reduce elevated total cholesterol, LDL cholesterol, apolipoprotein B (Apo-B) and triglycerides levels in various conditions such as familial hypercholesterolaemia, mixed dyslipidemia, hypertriglyceridaemia, dysbetalipoproteinaemia, coronary artery disease, and hypercholesterolemia associated with diabetes mellitus or renal transplantation.
DosageAndAdministration
- Adults: Usually 10 mg once daily; may be increased to max. 80 mg once daily if necessary.
- Child (10-18 years): Initially 10 mg once daily, increased to max. 20 mg once daily if necessary.
- It should be administered as a single dose at any time of the day, with or without food.
Pharmacology
- Absorption: Rapidly absorbed after oral administration; maximum plasma concentrations occur within 1 to 2 hours.
- Distribution: Mean volume of distribution is approximately 381 liters; 98% bound to plasma proteins.
- Metabolism: Extensively metabolized to ortho- and parahydroxylated derivatives and various beta-oxidation products.
- Excretion: Primarily eliminated in bile following hepatic and/or extra-hepatic metabolism.
Interaction
- Increased risk of myopathy with concurrent administration of cyclosporine, fibric acid derivatives, niacin, erythromycin, and azole antifungals.
- Plasma concentrations decreased when coadministered with antacid and increased when coadministered with oral contraceptives.
Contraindications
- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors

PrecautionsAndWarnings
Liver function tests should be performed before and periodically during treatment; therapy should be discontinued if markedly elevated CPK levels occur or myopathy is diagnosed or suspected.

- Geriatric: Plasma concentrations are higher in elderly subjects than in young adults.
- Pediatric: Pharmacokinetic data in the pediatric population are not available.
- RenalInsufficiency: No influence on plasma concentrations or LDL-C reduction.
- HepaticInsufficiency: Plasma concentrations are markedly increased in patients with chronic alcoholic liver disease.
OverdoseEffects
Specific treatment is not available; patient should be treated symptomatically and supportive measures instituted, as required.
