Indications
- Osteoarthritis

Pharmacology
Selective inhibition of COX-2, decrease in clinical signs and symptoms, decreased potential for GI toxicity, effects on platelet aggregation, inhibition of COX-2 without inhibition of COX-1

Interaction
- Interaction with oral anticoagulants
- Effects on diuretics, ACE inhibitors, and Angiotensin II Antagonists
- Concomitant use with acetylsalicylic acid for cardiovascular prophylaxis
- Potential interaction with Ciclosporin, tacrolimus, and lithium
Contraindications
- Hypersensitivity to the active substance or excipients
- Active peptic ulceration or active GI bleeding
- History of allergic-type reactions to NSAIDs or COX-2 inhibitors
- Pregnancy, lactation, severe hepatic dysfunction
- Estimated renal creatinine clearance <30 ml/min
- Children and adolescents under 16 years of age
- Inflammatory bowel disease, congestive heart failure, uncontrolled hypertension
- Established ischaemic heart disease, peripheral arterial disease, and/or cerebrovascular disease
Side Effects
- Palpitation, fatigue, influenza-like symptoms, ecchymosis
- Less commonly dry mouth, taste disturbance, mouth ulcer, appetite and weight change
- Atrial fibrillation, transient ischaemic attack, chest pain, flushing, cough
- Dyspnoea, epistaxis, mental acuity impaired, electrolyte disturbance, myalgia, arthralgia
- Rarely confusion and hallucinations
Pregnancy & Lactation
- Not recommended in women attempting to conceive
- Excreted in the milk of lactating rats
- Breastfeeding not recommended for women using the product
Precautions & Warnings
- Caution in patients at risk of developing gastrointestinal complications with NSAIDs
- Special consideration for patients with significant risk factors for cardiovascular events
- Possible reduction in prostaglandin formation and renal blood flow
- Caution in patients with history of cardiac failure, left ventricular dysfunction, hypertension
- Monitoring advised for patients with symptoms and/or signs suggesting liver dysfunction
- Discontinuation of product at the first appearance of specific signs
Overdose Effects
No significant toxicity observed with single or multiple doses up to certain levels
