Indications
- Symptomatic relief for Osteoarthritis (OA)

Pharmacology
- Orally active cyclooxygenase-2 (COX-2) specific inhibitor
- Reduces signs and symptoms of inflammation and pain
- Decreases potential for gastrointestinal (GI) toxicity
- Primarily responsible for the synthesis of prostanoid mediators of pain, inflammation, and fever
Dosage & Administration
- Recommended dose for Osteoarthritis: 30-60 mg once daily
- Recommended dose for Rheumatoid arthritis: 90 mg once daily
- Recommended dose for Ankylosing spondylitis: 90 mg once daily
- Recommended dose for Acute gouty arthritis: 120 mg once daily for 8 days
- Recommended dose for Postoperative dental surgery pain: 90 mg once daily
Interaction
- Increase in prothrombin time when used with oral anticoagulants
- Reduction in the effect of diuretics and other antihypertensive drugs when used with NSAIDs
- Concomitant use with acetylsalicylic acid at doses used for cardiovascular prophylaxis
- Potential increase in the nephrotoxic effect of ciclosporin or tacrolimus when used with NSAIDs
- Decrease in lithium renal excretion leading to increased lithium plasma levels when used with NSAIDs
Contraindications
- Hypersensitivity to the active substance or excipients
- Active peptic ulceration or active gastrointestinal (GI) bleeding
- Allergic-type reactions after taking acetylsalicylic acid or NSAIDs including COX-2 inhibitors
- Pregnancy and lactation
- Severe hepatic dysfunction
- Estimated renal creatinine clearance <30 ml/min
- Children and adolescents under 16 years
- Inflammatory bowel disease
- Congestive heart failure
- Persistently elevated blood pressure
- Established ischaemic heart disease, peripheral arterial disease, and/or cerebrovascular disease

Pregnancy & Lactation
- Not recommended in women attempting to conceive
- Unknown excretion in human milk

Precautions & Warnings
- Caution in patients at risk of developing a gastrointestinal complication with NSAIDs
- Caution in patients with significant risk factors for cardiovascular events
- May cause reduction in prostaglandin formation and impairment of renal function
- Caution in patients with a history of cardiac failure, left ventricular dysfunction, or hypertension
- Monitoring advised for patients with symptoms and signs suggesting liver dysfunction
- Discontinue at the first appearance of skin rash, mucosal lesions, or hypersensitivity
- May mask fever and other signs of inflammation
- Caution when co-administering with warfarin or other oral anticoagulants
Overdose Effects
- Administration of single doses up to 500 mg and multiple doses up to 150 mg/day did not result in significant toxicity
- Usual supportive measures to be employed in the event of overdose
