Description
- Atorvastatin is a selective inhibitor of HMG-CoA reductase
- Reduces total cholesterol, LDL cholesterol, apolipoprotein B (Apo-B) and triglycerides levels
- Treats diseases like heterozygous and homozygous familial hypercholesterolaemia, mixed dyslipidemia, hypertriglyceridaemia, dysbetalipoproteinaemia, coronary artery disease, hypercholesterolemia associated with or exacerbated by diabetes mellitus or renal transplantation
- Rapidly absorbed after oral administration, maximum plasma concentrations occur within 1 to 2 hours
Dosage and administration
- Usual dosage for adults: 10 mg once daily, can be increased to 80 mg once daily if necessary
- Usual dosage for children (10-18 years): Initially 10 mg once daily, increased to max. 20 mg once daily
Indications
- Adjunct to diet to reduce elevated total cholesterol, LDL cholesterol, apolipoprotein B (Apo-B) and triglycerides levels
Interaction
- Risk of myopathy increased with concurrent administration of cyclosporine, fibric acid derivatives, niacin (nicotinic acid), erythromycin, azole antifungals
Contraindications
- Hypersensitivity to any component of this medication
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors
Side effects
- Constipation, flatulence, dyspepsia, abdominal pain
- Infection, headache, back pain, rash, asthenia, arthralgia, myalgia
Pregnancy and lactation
- Contraindicated during pregnancy
- Not known whether excreted in human milk, contraindicated during breastfeeding
Precautions and warnings
- Liver function tests should be performed before the initiation of treatment and periodically thereafter
- Used with caution in patients who consume substantial quantities of alcohol or have a history of liver disease
Special populations
- Plasma concentrations higher in healthy elderly subjects (age 65 years) than in young adults
- No clinically significant difference in LDL-C reduction with Atorvast between men and women
Overdose effects
- Specific treatment is not available for atorvastatin overdose
- Liver function tests should be performed and serum CK levels should be monitored
