Pharmacology

  • Inhibition of leukotriene synthesis
  • Antibradykinin activity
  • Central Nervous System (CNS) suppression

Dosage

  • Anti-inflammatory dosage
  • Management of pain and primary dysmenorrhea
  • Recommended starting dose of 150 to 300 mg/day
  • Maintenance dosage of 100 to 200 mg/day
  • Maximum daily dose of 300 mg
  • Usual recommended dose of 25 to 50 mg every 6 to 8 hours

Administration

  • Oral administration
  • Should be taken with fluids
  • Preferably with food

Interaction

  • Not recommended drug associations
  • Increased risk of gastrointestinal ulceration and bleeding

Contraindications

  • Hypersensitivity reactions

  • Active peptic ulcer/hemorrhage
  • Gastrointestinal bleeding or perforation

  • Third trimester of pregnancy
  • Rectitis or history of proctorrhagia

Pregnancy & Lactation

  • Avoid use during the first and second trimester
  • Contraindicated during the last trimester
  • Not recommended in nursing mothers

Precautions & Warnings

  • Careful use in patients with gastrointestinal disease
  • Renal function monitoring required
  • Caution in patients with history of hypertension
  • Fluid retention and oedema reported
  • Increased risk of atrial fibrillation
  • Hyperkalemia may occur
  • Negative impact on female fertility

Use in Special Populations

  • Dosage reduction in the elderly
  • Careful monitoring in hepatic impairment
  • Dosage reduction in renal impairment

Overdose Effects

  • Reported symptoms include lethargy, drowsiness, nausea, vomiting
  • Gastric lavage may be recommended
  • Symptomatic and supportive treatment required
  • Hemodialysis may be useful in renal failure
