Indications
- Reduction of total cholesterol and LDL cholesterol in patients with familial hypercholesterolemia
- Treatment of mixed dyslipidemia and hypertriglyceridemia
- Reducing cardiac ischaemic events in patients with coronary artery disease
Pharmacology
- Selective inhibitor of HMG-CoA reductase
- Lowers plasma cholesterol and lipoprotein levels
- Increases number of hepatic LDL receptors for enhanced uptake and catabolism of LDL
Dosage & Administration
- Adult dosage range: 10 mg to 80 mg once daily
- Pediatric dosage range: 10 mg to 20 mg once daily
- Initial dose increased at intervals of at least 4 weeks
Interaction
- Increased myopathy risk with concurrent administration of cyclosporine, fibric acid derivatives, niacin, erythromycin, azole antifungals
- Plasma concentration alterations when coadministered with antacid, colestipol, digoxin, oral contraceptives, warfarin
Contraindications
- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations of serum transaminases

- No clinically significant difference in LDL-C reduction between men and women
- No dose adjustment in patients with renal dysfunction
