Indications
- Rheumatoid arthritis
- Degenerative joint diseases
- Musculoskeletal and joint disorders such as tendinitis
- Pain, regardless of the origin, such as dental pain, headache and primary dysmenorrhea
Pharmacology
- Non-steroidal anti-inflammatory drug (NSAID)
- Analgesic
- Antipyretic
- Inhibition of prostaglandin synthesis
- Stabilization of lysosomal membranes
- Inhibits leukotriene synthesis
- Antibradykinin activity
- Inhibition of prostaglandin synthesis in the CNS
- Rapid and almost complete absorption from the GI tract
- Approximately 99% bound to plasma protein
- Rapid and extensive metabolism in the liver
- Excretion in urine and faeces
Dosage
- Anti-inflammatory dosage: 150 to 300 mg/day in 3 divided doses
- Once the maintenance dosage is established, consideration of a twice daily dose regimen
- Maximum daily dose: 300 mg
- Management of pain and primary dysmenorrhea: 25 to 50 mg every 6 to 8 hours, not to exceed 300 mg per day
Administration
- Oral forms should be taken with fluids, preferably with food
Interactions
- Not recommended drug associations with other NSAIDs and high dose salicylates
- Increased risk of bleeding with anticoagulants
- Interactions with lithium, methotrexate, and other medicinal products and therapeutic categories
Contraindications
- Hypersensitivity reactions to ketoprofen, ASA or other NSAIDs
- Severe heart failure
- Active or history of peptic ulcer/hemorrhage
- Severe hepatic or renal insufficiency
- Third trimester of pregnancy

Pregnancy & Lactation
- Avoid use during first and second trimester of pregnancy
- Contraindicated during the third trimester
- Not recommended in nursing mothers
Precautions & Warnings
- NSAIDs should be given with care to patients with a history of gastrointestinal disease
- Careful monitoring of renal function in specific patient groups
- Caution in patients with hypertension and congestive heart failure
- Increased risk of atrial fibrillation and hyperkalemia
- Masking of signs of infection progression
- Periodic evaluation of liver function tests
- Consideration of visual disturbances and female fertility effects
Use in Special Populations
- Elderly: reduced initial dosage and maintenance on minimal effective dose
- Hepatic impairment: careful monitoring and minimal effective daily dosage
- Renal impairment: reduced initial dosage and maintenance on minimal effective dose
Overdose Effects
- Reported cases with symptoms such as lethargy, nausea, vomiting, and epigastric pain
- No specific antidotes, symptomatic and supportive treatment recommended
- Consideration of gastric lavage and hemodialysis if necessary
