Description
- Napoxen is indicated for the relief of sign and symptoms of rheumatoid arthritis, osteoarthritis, ankylosing spondylitis, juvenile arthritis, tendonitis, bursitis, acute gout, dysmenorrhea, and pain.
- It is also indicated for the management of primary dysmenorrhea & pain.
- Naproxen exerts its clinical effects by blocking COX-1 and COX-2 enzymes leading to decreased prostaglandin synthesis.
- Naproxen oral preparations should be taken with water preferably after meals.
- ACE inhibitors diminish the antihypertensive effect of ACE inhibitors.
- Naproxen is contraindicated in patients with known hypersensitivity to Naproxen.
- Most frequently reported side effects include gastrointestinal, central nervous system, dermatological, cardiovascular, and other effects.
- Naproxen causes a delay in parturition in animals and has been associated with premature closure of the ductus arteriosus and severe pulmonary hypertension in infants born to mothers taking Naproxen.
- As with other non-steroidal anti-inflammatory drugs, long-term administration of naproxen to animal has resulted in renal papillary necrosis and other abnormal renal pathology.

Mode of Action
- Blocks COX-1 and COX-2 enzymes
- Decreases prostaglandin synthesis
Dosage & Administration
- Naproxen Enteric Coated Tablet for adults: 500-1000 mg per day taken in two divided doses at 12-hour intervals.
- For acute gout: 750 mg should be given initially, then 250 mg every 8 hours until the attack has passed.
- Naproxen Suspension for juvenile rheumatoid arthritis: The usual dose for children over 2 years is 10 mg/kg/day given as two divided doses at 12-hour intervals.
- Naproxen Gel is to be applied 2-6 times a day as required and is not recommended for use in children.
Interaction
- ACE inhibitors diminish the antihypertensive effect of ACE inhibitors.
- Antacids & Sucralfate delay the absorption of Napoxen.
- Aspirin increases adverse effects.
- Diuretics reduce the natriuretic effect of Furosemide and Thiazides.
- Methotrexate enhances the toxicity of Methotrexate.
- Warfarin increases the risk of GI bleeding.
- Selective Serotonin Reuptake Inhibitors (SSRI) increase the risk of GI bleeding.
Contraindications
- Hypersensitivity to Naproxen
- Asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs
- Perioperative pain in the setting of coronary artery bypass graft (CABG) surgery
