Pharmacology
- Mechanism of action involves blocking COX-1 and COX-2 enzymes
- Decreased prostaglandin synthesis

Dosage & Administration
- Oral administration with water after meals

Interaction
- Diminished antihypertensive effect with ACE inhibitors
- Delayed absorption with antacids & sucralfate
- Increased adverse effects with aspirin
- Reduced natriuretic effect of diuretics
- Enhanced toxicity of methotrexate
- Increased risk of GI bleeding with warfarin and selective serotonin reuptake inhibitors (SSRI)
Contraindications
- Known hypersensitivity to naproxen
- Asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs
- Treatment of perioperative pain in the setting of coronary artery bypass graft (CABG) surgery
Side Effects
- Gastrointestinal side effects (heartburn, abdominal pain, nausea, diarrhea, dyspepsia)
- Central nervous system side effects (headache, vertigo, drowsiness)
- Dermatological side effects (pruritus, purpura)
- Cardiovascular side effects (edema, palpitation)
- Other side effects (visual and hearing disturbances)
Pregnancy & Lactation
- Adverse effects on the fetus
- Delayed parturition and potential risks to the fetus
- Excretion in small amounts in breast milk
- Recommendation to avoid breastfeeding during therapy
Precautions & Warnings
- Renal effects including abnormal renal pathology and nephritis
- Monitoring of serum creatinine and/or creatinine clearance
- Caution in patients with significantly impaired renal function
- Caution in patients with creatinine clearance of less than 20ml/min
- Reports of moderate to severe jaundice attributed to Naxin
