Indications
- Relief of signs & symptoms of Osteoarthritis
- Relief of signs & symptoms of Rheumatoid arthritis
- Relief of signs & symptoms of Ankylosing spondylitis
- Decrease the risk of developing gastric ulcers in patients at risk of developing NSAID-associated gastric ulcers
Pharmacology
- Consists of an immediate release Esomeprazole Magnesium layer
- Consists of an enteric-coated Naproxen core
- Inhibition of prostaglandin synthesis by Naproxen
- Suppression of gastric acid secretion by Esomeprazole
Dosage
- Use the lowest effective dose for the shortest duration
- Dosage for Rheumatoid Arthritis, Osteoarthritis and Ankylosing Spondylitis
- Dosage for Juvenile Idiopathic Arthritis in Adolescent Patients 12 Years of Age & Older
Administration
- Tablet should not be split, chewed, crushed or dissolved
- To be taken at least 30 minutes before meals
Interaction
- Concomitant use of NSAIDs may reduce the antihypertensive effect of ACE inhibitors, diuretics & beta-blockers
- Concomitant use of the tablet and warfarin may result in increased risk of a bleeding complication
- Inhibition of gastric acid secretion by Esomeprazole may interfere with the absorption of certain drugs
Contraindications
- Known hypersensitivity to any component of the tablet or substituted benzimidazoles
- History of asthmay urticaria or other allergic-type reactions after taking aspirin or other NSAIDs
- Use during the peri-operative period in the setting of coronary artery bypass graft (CABG) surgery
Side Effects
- Naproxen and Esomeprazole tablet has been shown to significantly decrease the occurrence of gastric ulcers
- NSAID use may be associated with a small increased risk of arterial thrombotic events
- Gastrointestinal adverse events may occur with NSAID use
Pregnancy & Lactation
- Naproxen and Esomeprazole tablet should not be given during the first and second trimester of pregnancy unless the potential benefit outweighs the potential risk
- Naproxen and Esomeprazole tablet is contraindicated during the third trimester of pregnancy
- Naproxen and Esomeprazole tablet should not be used during breastfeeding
Precautions & Warnings
- The combination of Naproxen and Esomeprazole tablet and NSAIDs including cyclooxygenase-2 selective inhibitors should be avoided
- Undesirable effects may be minimized by using the lowest effective dose for the shortest duration necessary
- Risk-factors to develop NSAID related gastro-intestinal complications include high age, concomitant use of anticoagulants, corticosteroids, other NSAIDs including low-dose acetylsalicylic acid, debilitating cardiovascular disease
Use in Special Populations
- Use caution when high doses are required in elderly patients
- Naproxen-containing products are not recommended for use in patients with moderate to severe renal impairment
- Monitor patients with mild to moderate hepatic impairment closely
