Indications

  • Relief of Osteoarthritis
  • Relief of Rheumatoid arthritis

Pharmacology

  • Consists of immediate release Esomeprazole Magnesium layer & enteric-coated Naproxen core
  • Esomeprazole inhibits gastric acid secretion
  • Naproxen inhibits prostaglandin synthesis

Dosage

  • Consider potential benefits & risks
  • Use lowest effective dose for shortest duration
  • Adult dosage: 375mg naproxen/20mg esomeprazole or 500mg naproxen/20mg esomeprazole
  • Adolescent dosage: Varies by weight

Administration

  • Do not split, chew, crush or dissolve tablet
  • Take at least 30 minutes before meals

Interaction

  • Concomitant use with NSAIDs may reduce antihypertensive effect of ACE inhibitors, diuretics & beta-blockers
  • Concomitant use with warfarin may increase risk of bleeding
  • Interference with absorption of drugs like Ketoconazole, iron salts, digoxin

Contraindications

  • Hypersensitivity to any component of the tablet
  • History of asthmay urticaria or other allergic-type reactions after taking aspirin or other NSAIDs
  • Use during peri-operative period in the setting of coronary artery bypass graft (CABG) surgery

Side Effects

  • Gastrointestinal side effects reduction
  • Potential risk of arterial thrombotic events
  • Possible adverse events include oedema, hypertension, cardiac failure, peptic ulcers, nausea, vomiting, diarrhoea, etc.

Pregnancy & Lactation

  • Use not recommended during third trimester of pregnancy
  • Use not recommended during breastfeeding
  • May impair female fertility

Precautions & Warnings

  • Avoid use with NSAIDs including cyclooxygenase-2 selective inhibitors
  • Use with low dose acetylsalicylic acid recommended
  • Regular surveillance for long-term treatment patients

Use in Special Populations

  • Caution in elderly patients
  • Not recommended for patients with moderate to severe renal impairment
  • Use with caution in patients with mild to moderate hepatic impairment

Overdose Effects

  • Symptoms related to Naproxen and esomeprazole overdose
