Indications
- Acute and chronic rheumatoid arthritis
- Systemic Lupus Erythematosus (SLE)
- Malaria
Pharmacology
Acts as an immunosuppressant by inhibiting production of rheumatoid and acute phase reactants, accumulates in white blood cells, stabilizes lysosomal membranes, and inhibits the activity of many enzymes. It also disrupts cell walls of infected red blood cells and kills developing parasites.
Dosage & Administration
- Acute and chronic rheumatoid arthritis: 400 to 600 mg daily, can be reduced to 50% after good response (usually 4 to 8 weeks)
- Systemic Lupus Erythematosus (SLE): 400 mg once or twice daily for several weeks or months, maintenance dose is 200 to 400 mg daily
- Malaria:
- Adults: Initial dose of 800 mg, followed by 400 mg in 6-8 hours and 400 mg on each of two consecutive days
- Children: Total dose representing 25 mg/kg is administered in 3 days following specific doses based on weight

Pregnancy & Lactation
Should be used only if clearly needed during pregnancy, small amounts found in breast milk
Precautions & Warnings
Children are especially sensitive, keep out of reach of children, opthalmologic examination required every 12 months
Overdose Effects
Symptoms include headache, drowsiness, visual disturbances, cardiovascular collapse, convulsions, and respiratory and cardiac arrest. Gastric lavage recommended until stomach is emptied.
