Indications
- Chronic management of hyperuricemia in patients with gout
Pharmacology
- Non-purine, selective xanthine oxidase (XO) inhibitor
Dosage & Administration
- Recommended starting dose of 40 mg tablet once daily
- Dose adjustment for tumor lysis syndrome
- Gout flare management
- No dose adjustment for renal or hepatic impairment
- Safety and effectiveness not established in pediatric patients
Interaction
- Contraindicated with mercaptopurine/azathioprine/theophylline
- Co-administration with rosiglitazone or CYP2C8 substrates
- Effect on metabolism by naproxen and other inhibitors of glucuronidation
- Effects of potent inducers of glucuronidation
- Co-administration with colchicine/indometacin/hydrochlorothiazide/warfarin
- Interaction with antacids
Contraindications
- Contraindicated in patients being treated with azathioprine, mercaptopurine, or theophylline
Side Effects
- Commonly reported adverse reactions including gout flares, liver function abnormalities, diarrhea, nausea, headache, rash, and edema
- Rare serious hypersensitivity reactions observed
Pregnancy & Lactation
- Pregnancy Category C
- Caution advised during breastfeeding
Precautions & Warnings
- Increase in gout flares during initiation of treatment
- Higher rate of cardiovascular thromboembolic events observed than allopurinol
- Monitoring for signs and symptoms of MI and stroke
- Transaminase elevations observed in Eburic-treated patients
- Periodic liver function tests monitoring
Overdose Effects
- Studied in healthy subjects in doses up to 300 mg daily for seven days without evidence of dose-limiting toxicities
