Indications
- Chronic management of hyperuricemia in patients with gout
Pharmacology
- Non-purine, selective xanthine oxidase (XO) inhibitor
- Decreases serum uric acid level by inhibiting xanthine oxidase
Dosage & Administration
- Recommended starting dose of Febuxostat 40 mg tablet
- Additional dosage guidelines for specific conditions such as Tumor Lysis Syndrome and Gout Flares
- No dose adjustment required for patients with mild to moderate renal or hepatic impairment
- Safety and effectiveness not established in pediatric patients under 18 years of age
Interaction
- Contraindicated with mercaptopurine, azathioprine, or theophylline
- No dose adjustment required for co-administration with rosiglitazone, naproxen, colchicine, indomethacin, hydrochlorothiazide, warfarin, or antacids
- Possible effect on UGT enzymes and potential interactions with inducers or inhibitors of glucuronidation
Contraindications
- Contraindicated in patients being treated with azathioprine, mercaptopurine, or theophylline
Side Effects
- Most commonly reported adverse reactions include gout flares, liver function abnormalities, diarrhea, nausea, headache, rash, and edema
- Rare serious hypersensitivity reactions observed

- Caution advised during breastfeeding
Precautions & Warnings
- Increased gout flares observed during initiation of anti-hyperuricemic agents
- Higher rate of cardiovascular thromboembolic events observed in clinical trials
- Transaminase elevations observed, monitor liver function tests periodically
Overdose Effects
- Studied in healthy subjects in doses up to 300 mg daily for seven days without evidence of dose-limiting toxicities
