Indications
- Spasticity resulting from multiple sclerosis
- Flexor spasms and concomitant pain, clonus and muscular rigidity
- Skeletal muscle spasm resulting from rheumatic disorders
- Spinal cord injuries and other spinal cord diseases
- Cerebrovascular accidents or neoplastic or degenerative brain disease
Pharmacology
- Inhibition of monosynaptic and polysynaptic reflexes
- Stimulation of GABA receptors
- Antinociceptive effect
Dosage & Administration
- 5 mg 3 times daily for adults & children over 10 years
- Gradually increased dosage
- Maximum daily dosage of 100 mg
- Adjusted dosage for children under 10 years based on age
Interaction
- Increased sedation when taken with CNS agents, alcohol, or synthetic opiates
- Likely blood pressure fall when combined with antihypertensives
- Potentiation of effects when taken with tricyclic antidepressants
- Reports of mental confusion and hallucinations when combined with levodopa and carbidopa
- Increased CNS depressant effects when combined with MAO inhibitors
- Caution advised when combined with magnesium sulphate or neuromuscular blocking agents
Contraindications
- Hypersensitivity to any component of the product
Side Effects
- Transient drowsiness
- Daytime sedation
- Dizziness
- Weakness
- Fatigue
- Various CNS effects including headache, insomnia, euphoria, depression, seizures
- Cardiovascular effects like hypotension, dyspnoea, palpitation
- Gastrointestinal effects including nausea, constipation, dry mouth
- Genitourinary effects like urinary frequency, enuresis, urinary retention
- Other effects such as rash, pruritus, weight gain, hepatic function disorders
Pregnancy & Lactation
- Pregnancy category B3
- Use only when benefits outweigh potential hazards
- Negligible excretion in breast milk
Precautions & Warnings
- Lower doses for patients with impaired renal function or chronic haemodialysis
- Cautious use in patients with psychotic disorders, schizophrenia, depressive or manic disorders
- Possible lowering of convulsion threshold in patients with epilepsy
- Caution in patients with peptic ulcers, cerebrovascular diseases, or organ failure
- Monitoring of respiratory and cardiovascular function especially in cardiopulmonary disease
- Improvement in neurogenic disturbances affecting bladder emptying
- Caution in patients with preexisting sphincter hypertonia
- No significant benefit for stroke patients
Overdose Effects
- Gastric lavage in case of severe overdose
