Indications
- Spasticity resulting from multiple sclerosis
- Flexor spasms and concomitant pain, clonus and muscular rigidity
- Skeletal muscle spasm resulting from rheumatic disorders
- Spinal cord injuries and other spinal cord diseases
- Cerebrovascular accidents or neoplastic or degenerative brain disease
Pharmacology
- Baclofen inhibits monosynaptic and polysynaptic reflexes
- Stimulates GABA receptors
- Inhibits release of glutamate and aspartate
- Produces CNS depression
- Exerts antinociceptive effect
Dosage and Administration
- Adult & children over 10 years: 5 mg 3 times daily, max. 100 mg daily
- Children <10 years: Treatment started with 2.5 mg 4 times daily, raised according to requirement
- Doses adjusted based on age
Interaction
- Increased sedation with agents acting on the central nervous system, alcohol or synthetic opiates
- Likely increase in fall in blood pressure with antihypertensives
- Potentiation of effects with tricyclic antidepressants
- Reports of mental confusion, hallucinations, headaches, nausea and agitation with levodopa/carbidopa and Flexilax
- CNS depressant effects with MAO inhibitors
- Potential synergistic effect with magnesium sulphate or other neuromuscular blocking agents
Contraindications
- Hypersensitivity to any component of the product
Side Effects
- Transient drowsiness
- Daytime sedation
- Dizziness
- Weakness
- Fatigue
- Adverse reactions in Central Nervous System, Cardiovascular, Gastrointestinal, Genitourinary, and other systems
Pregnancy and Lactation
- Pregnancy category B3
- Safe use during pregnancy not established
- Excretion in breast milk with quantities too small to cause undesirable effects on the infant
Precautions and Warnings
- Lower doses for patients with impaired renal function or undergoing chronic haemodialysis
- Cautious treatment for patients with psychotic disorders, schizophrenia, depressive or manic disorders, or confusional states
- Potential lower convulsion threshold in patients with epilepsy and muscle spasticity
- Caution in patients with peptic ulcers, cerebrovascular diseases, or hepatic, renal or respiratory failure
- Careful monitoring of respiratory and cardiovascular function
- Improvement in neurogenic disturbances affecting bladder emptying
- Poor tolerance in stroke patients
- Periodic laboratory tests for patients with hepatic diseases or diabetes mellitus
Overdose Effects
- Importance of gastric lavage in cases of severe overdose
