Indications
- spasticity resulting from multiple sclerosis
- flexor spasms and concomitant pain, clonus and muscular rigidity
- skeletal muscle spasm resulting from rheumatic disorders
- spinal cord injuries and other spinal cord diseases
- cerebrovascular accidents or neoplastic or degenerative brain disease
Pharmacology
- inhibits both monosynaptic and polysynaptic reflexes at the spinal level
- stimulates GABA-B receptors
- inhibits release of glutamate and aspartate
- antinociceptive effect
- CNS depression
Dosage & Administration
- 5 mg 3 times daily for adults & children over 10 years
- gradually increased dosage up to max. 100 mg daily
- dosage for children under 10 years based on age and weight
Interaction
- increased sedation when taken with CNS acting agents, alcohol, or synthetic opiates
- increased fall in blood pressure when combined with antihypertensives
- potentiation of pharmacological effects when used with tricyclic antidepressants
- reports of mental confusion, hallucinations, headaches, nausea, and agitation when taken with levodopa and carbidopa
Contraindications
- hypersensitivity to any component of this product

- fatigue
- adverse reactions including CNS, cardiovascular, gastrointestinal, genitourinary, and other systemic effects
Pregnancy & Lactation
- Pregnancy category B3
- safe use during pregnancy not established
- excretion in breast milk but quantities expected to have no undesirable effects on the infant
Precautions & Warnings
- lower doses for patients with impaired renal function or undergoing chronic haemodialysis
- treatment caution for patients with psychotic disorders, schizophrenia, depressive or manic disorders
- monitoring for exacerbations of disorders, seizures after the cessation of therapy or overdose
- special caution in patients with peptic ulcers, cerebrovascular diseases, or hepatic, renal, or respiratory failure
- respiratory and cardiovascular function monitoring
- caution in patients with preexisting sphincter hypertonia
- limited benefit and poor tolerance in stroke patients
- periodic laboratory tests for patients with hepatic diseases or diabetes mellitus
Overdose Effects
- importance of gastric lavage in severe overdose
