Indications
- Reduction of total cholesterol and LDL cholesterol levels
- Treatment of hypercholesterolaemia and dyslipidemia
- Prevention of cardiovascular events
- Reduce cardiac ischaemic events
- Associated with diabetes mellitus or renal transplantation
Pharmacology
- Selective inhibitor of HMG-CoA reductase
- Lowers plasma cholesterol and lipoprotein levels
- Increases hepatic LDL receptors for enhanced uptake and catabolism of LDL
- Rapidly absorbed after oral administration
DosageAndAdministration
- Adults dosage range: 10 to 80 mg once daily
- Child dosage range: 10 to 20 mg once daily
- Initiate on a standard cholesterol-lowering diet
- Lipid levels should be analyzed within 2 to 4 weeks and dosage adjusted accordingly
Interactions
- Increased risk of myopathy with concurrent administration of cyclosporine, fibric acid derivatives, niacin, erythromycin, azole antifungals
- Decreased plasma concentrations when coadministered with antacid or colestipol
- Monitoring required with coadministration of digoxin, erythromycin, oral contraceptives, and warfarin

- Hypersensitivity to any component
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior HMG-CoA reductase inhibitors

- Discontinue therapy if markedly elevated CPK levels occur or myopathy is diagnosed or suspected

- Higher plasma concentrations in healthy elderly subjects
- No clinically significant difference in LDL-C reduction between men and women
- Renal disease has no influence on plasma concentrations or LDL-C reduction

- Liver function tests and serum CK levels should be monitored
