Pharmacology

  • Selective inhibitor of HMG-CoA reductase
  • Lowers plasma cholesterol and lipoprotein levels
  • Increases hepatic LDL receptors for enhanced uptake of LDL
  • Rapidly absorbed after oral administration
  • Maximum plasma concentrations occur within 1 to 2 hours

Dosage & Administration

  • Recommended starting dose of 10 or 20 mg once daily
  • Maximum dose of 80 mg once daily
  • Can be administered as a single dose at any time of the day
  • Dose adjustment based on lipid levels analysis

Interaction

  • Increased risk of myopathy with concurrent administration of certain drugs
  • Plasma concentrations affected by coadministration with antacid and colestipol
  • Altered levels of digoxin and oral contraceptives when coadministered
  • No clinically significant effect on prothrombin time when administered with warfarin

Contraindications

  • Hypersensitivity to any component of the medication
  • Active liver disease or unexplained persistent elevations of serum transaminases
  • History of serious adverse reaction to HMG-CoA reductase inhibitors

  • Higher plasma concentrations in healthy elderly subjects
  • Greater degree of LDL-lowering in elderly patients
  • Pharmacokinetic data not available in pediatric population
  • No clinically significant difference in LDL-C reduction between men and women

Overdose Effects
