Indications
- Adjunct to diet to reduce elevated total cholesterol levels

Pharmacology
- Selective inhibitor of HMG-CoA reductase
- Lowers plasma cholesterol and lipoprotein levels
- Increases hepatic LDL receptors for enhanced uptake of LDL
- Rapidly absorbed after oral administration
- Maximum plasma concentrations occur within 1 to 2 hours
Dosage & Administration
- Recommended starting dose of 10 or 20 mg once daily
- Maximum dose of 80 mg once daily
- Can be administered as a single dose at any time of the day
- Dose adjustment based on lipid levels analysis
Interaction
- Increased risk of myopathy with concurrent administration of certain drugs
- Plasma concentrations affected by coadministration with antacid and colestipol
- Altered levels of digoxin and oral contraceptives when coadministered
- No clinically significant effect on prothrombin time when administered with warfarin
Contraindications
- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to HMG-CoA reductase inhibitors

- Caution in patients with substantial alcohol consumption or liver disease
Use in Special Populations
- Higher plasma concentrations in healthy elderly subjects
- Greater degree of LDL-lowering in elderly patients
- Pharmacokinetic data not available in pediatric population
- No clinically significant difference in LDL-C reduction between men and women
Overdose Effects
