Description
- Lipex is indicated as an adjunct to diet to reduce elevated total cholesterol, LDL cholesterol, apolipoprotein B (Apo-B) and triglycerides levels in various diseases when response to diet and other non-pharmacological measures is inadequate.
Dosage Instructions
- Adults: Usually 10 mg once daily; may be increased at intervals of at least 4 weeks to max. 80 mg once daily.
Pharmacological Action
- Selective inhibitor of HMG-CoA reductase
- Lowers plasma cholesterol and lipoprotein levels
- Increases the number of hepatic LDL receptors
Absorption
- Rapidly absorbed after oral administration; maximum plasma concentrations occur within 1 to 2 hours
Distribution
- Mean volume of distribution of Atorvastatin is approximately 381 liters
- Atorvastatin is 98% bound to plasma proteins
Metabolism
- Extensively metabolized to ortho- and parahydroxylated derivatives and various beta-oxidation products
Excretion
- Atorvastatin and its metabolites are eliminated primarily in bile following hepatic and/or extra-hepatic metabolism
Interaction
- Increased risk of myopathy with concurrent administration of cyclosporine, fibric acid derivatives, niacin, erythromycin, azole antifungals
- Altered plasma concentrations with antacid, colestipol, digoxin, erythromycin, oral contraceptives
Contraindications
- Hypersensitivity to any component of this medication
- Active liver disease or unexplained persistent elevations of serum transaminases

Pregnancy & Lactation
- Contraindicated during pregnancy

Precautions & Warnings
- Liver function tests should be performed before the initiation of treatment and periodically thereafter
- Discontinue therapy if markedly elevated CPK levels occur or myopathy is diagnosed or suspected.
Use in Special Populations
- Plasma concentrations of Lipex are higher in healthy elderly subjects than in young adults
- No clinically significant difference in LDL-C reduction with Lipex between men and women
Overdose Effects
- Specific treatment is not available for atorvastatin overdose
- Liver function tests should be performed and serum CK levels should be monitored
