Dosage

Metabolism

  • Extensively metabolized to ortho- and parahydroxylated derivatives
  • Metabolism by cytochrome P450 3A4

Excretion

  • Eliminated primarily in bile
  • Plasma elimination half-life approximately 14 hours

Interaction

  • Risk of myopathy with concurrent use of certain drugs
  • Effects on plasma concentrations when coadministered with antacid, colestipol, digoxin, etc.

  • Hypersensitivity to any component

  • History of serious adverse reaction to HMG-CoA reductase inhibitors

  • No clinically significant difference in LDL-C reduction between men and women
  • No dose adjustment necessary for renal dysfunction
