- Reducing elevated total cholesterol
- Reducing LDL cholesterol
- Reducing apolipoprotein B levels

Mode of Action
- Selective inhibitor of HMG-CoA reductase
- Lowers plasma cholesterol and lipoprotein levels
- Increases hepatic LDL receptors for enhanced uptake of LDL
Pharmacokinetics
- Rapid absorption after oral administration
- Maximum plasma concentrations within 1 to 2 hours
- Extensively metabolized to active metabolites
Dosage and Administration
- Adult dosage usually 10 mg once daily
- Maximum dosage of 80 mg once daily
- Pediatric dosage of 10-20 mg once daily for 10-18 years
Interactions
- Increased risk of myopathy with concurrent use of certain drugs
- Altered plasma concentrations with antacids and colestipol
- Increased plasma digoxin concentrations when coadministered with atorvastatin
Contraindications
- Hypersensitivity to any component
- Active liver disease
- Unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior HMG-CoA reductase inhibitors

- Higher plasma concentrations in elderly subjects
- No clinically significant difference in LDL-C reduction between men and women
- Renal disease and hemodialysis do not influence plasma concentrations or LDL-C reduction
